Objective: 

This laboratory course aims to develop practical skills in fundamental microbiological techniques, including aseptic handling, isolation, identification, enumeration, and preservation of microbes and mutants from various samples. Students will also analyze microbial metabolic capabilities, evaluate susceptibility to antibiotics and disinfectants, and apply Mendelian principles to study gene interactions.

Laboratory II: Microbiology and Genetics

(Practical)

CO43:Design experiments for isolation,identification, preservation and enumeration of microbes present in a sample. CO44: Evaluate the metabolic capabilities of microbes and determine their susceptibility to antibiotics and disinfectants.

CO45:Design experiments to isolate mutants and apply Mendelian inheritance principles and gene interaction models to analyze genetic variations.

CO46: Identify and illustrate the various elements of microbiology

CO47: Discuss and defend the concepts of microbiology and illustrate the exercises performed with appropriate methods and outcomes and maintain proper documentation of the same.

CO48: Contribute effectively in course-specific interaction

Experiments based on:

Basic Microbiological Techniques

  1. Sterilization, disinfection and safety in microbiological laboratory.
  2. Preparation of media for cultivation of bacteria.
  3. Isolation of bacteria in pure culture by streak plate method.
  4. Maintenance of stock cultures: slants, stabs and glycerol stock cultures
  5. Study of colony and growth characteristics of some common bacteria: Bacillus, E. coli,Staphylococcus, Streptococcus, etc.
  6. Preparation of bacterial smear and Staining methods: Negative, Gram’s staining, endospore staining.
  7. Enumeration of bacteria: standard plate count.
  8. Biochemical characterization of microbes.
  9. Isolation and identification of bacteria from soil/water samples.
  10. Antimicrobial sensitivity test and demonstration of drug resistance.
  11. Determination of phenol co-efficient of antimicrobial agents.
  12. Determination of Minimum Inhibitory Concentration (MIC)
  13. Isolation and identification of Rhizobium from root nodules of leguminous plants.
  14. Isolation and identification of Azotobacter from soil.

Virology, Mycology and Phycology

  1. Study of various symptoms produced in plants due to virus infection.
  2. Study of viral diseases of plants/animals/human (Specimen/photographs)
  3. Different type of viruses (Photographs/sketches).
  4. Titration of Phages
  5. Isolation and identification of fungi.
  6. Study of permanent slides of algae and fungi

Genetics

  1. Isolation of antibiotic-resistant microorganisms by replica plating.
  2. Isolation of antibiotic-resistant microorganisms by Gradient plate technique
  3. Simple problems based on Mendel’s laws and gene interactions.
  4. Any other practical based on theory syllabus.
