  • Brief history of Plant Tissue Culture (Contribution of Indian Scientist), Introduction to in vitro Methods. Terms and definition.
  • Plant Tissue Culture media.Concept of totipotency Embryo culture, embryo rescue after hybridization and its applications.
  • Introduction to the processes of embryogenesis and organogenesis and their practical applications.

  • Micropropagation: axillary bud, shoot- tip and meristem culture,anther culture.
  • Haploids and their applications. SomaCOnal variations and applications. In vitro pollination and fertilization. Endosperm culture and production of triploids.Single cell suspension cultures.

·         Introduction to protoplast isolation: Principles and applications.

·         Testing of viability of isolated protoplasts

  • Use of markers for selection of hybrid cells.
  • Practical applications of somatic hybridization (Hybrids vs. Cybrids)
  • Use of plant cell and tissue culture for genetic manipulation of plants. Marker assisted plant breeding.

  • Methods of gene transfer: Direct and Agarobacterium mediated. Introduction of A. tumefaciens and A. rhizogenes (Monocots vs. Dicots). Formation of hairy root culture. Practical application of genetic transformation .
