Objective: 

The course will develop in depth understanding of fermenter design, operation, process parameters and recovery methods. In addition, it will also acquaint them to tools of genetic engineering and gene transfer techniques.

Bioreactors and Fermentation Process

Bioreactors:  Basic Design, Conventional and Specialized bioreactors (pulsed, fluidized, photo bioreactors), Types of fermentation processes batch, fed batch and continuous cultures. Primary and Secondary screening techniques.

Unit II: 
Media Production and Control of Fermentation Parameters

Raw materials used in media production, inoculum preparation, industrial sterilization, Probes for process monitoring and control of fermentation parameters (temperature, pH, dissolved oxygen, foam).

Unit III: 
Principles and Operation of Downstream Processing

Cell disruption, filtration, centrifugation, liquid-liquid extraction, solvent extraction (distillation), preparative ion exchange chromatography, drying.

Unit IV: 
Recombinant DNA Technology-I

Gene cloning vectors- Plasmids, Bacteriophages, Cosmids, phagemids, YAC, BAC; Enzymes used in gene manipulation- Restriction endonucleases, DNA polymerase, Polynucleotide kinase, ligase, Terminal deoxynucleotidyl transferase.

Unit V: 
Recombinant DNA Technology- II

Methods of gene transfer, Making and applications of genomic and c-DNA libraries; Blotting Techniques- Southern, Northern and Western Blotting.
